Ribonuclease H (RNase H) is an endoribonuclease that specifically hydrolyzes the phosphodiester bonds of RNA which is hybridized to DNA (RNA-DNA hybrids).

Applications
- Degradation of mRNA - prior to synthesis of second strand cDNA
- Degradation of RNA after first strand cDNA synthesis - RT-PCR and RT-QPCR
The RNAse H can be inactivated by heating at 65 °C for 10 min.
