Media without phenol red
Cell culture media usually contain the dye phenol red as a pH indicator. During the cultivation of cells, the pH value of the medium shifts towards the acidic range over time. By changing color from red to yellow, the indicator shows the increasing acidification of the medium. If the pH value shifts into the alkaline range, the color changes from red to violet. This color change allows easy visual control of the pH value of the medium and thus the growth conditions of the cultivated cells.
In some cases, however, the presence of phenol red is undesirable. Especially in the culture of primary cells, the use of media with phenol red can lead to the formation of artifacts under certain circumstances. The presence of phenol red can also interfere with or make it impossible to perform chromogenic bioassays.
