RPMI 1640 was developed at Roswell Park Memorial Institute (RPMI) in 1966 to support lymphoblastoid cells in suspension cultures. The formulation of RPMI 1640 is clearly different from other cell culture media. It contains the reducing reagent glutathione and high vitamin concentrations. As a result, RPMI 1640 also has a wide range of applications for other cell types such as PBMC, MCF-7, HeLa, Jurkat, astrocytes, PC12 and carcinomas. For some cell lines it may be necessary to supplement RPMI 1640 with fetal calf serum. However, it has been shown that the cell culture medium is also suitable for cultivating various cell lines without serum.
