Our BioColl® separation solution contains neutral, highly cross-linked polysucrose with an average molecular weight of 400,000 Da. This hydrophilic polymer can be used to produce aqueous solutions with different densities (up to about 1.2 g/ml), which are used to isolate cells or subcellular particles. The appropriate density of the BioColl® separation solution is selected depending on the particle type. A density of 1.1 g/ml is ideal for the isolation of human granulocytes. For optimum pH and osmolality values, the BioColl® separation solution should be adjusted with an acid (preferably amidotrizoic acid (ATS)) and sodium hydroxide solution.
In order to specifically isolate granulocytes, a double polysucrose gradient is required. For this purpose, BioColl® for lymphocytes (BS.L6115) is layered over BioColl® for granulocytes (BS.L6155). Lymphocytes, thrombocytes and monocytes do not penetrate the Biocoll® layer for lymphocytes (density 1.077 g/ml) due to their lower density and can therefore be easily removed. Due to their higher density, granulocytes pass through the BioColl® layer for lymphocytes (density 1.077 g/ml), but not the BioColl® layer for granulocytes (density 1.1 g/ml). After centrifugation, they are located between the two BioColl® layers. The cells can be easily harvested by careful pipetting.

This physical process is based on the separation of macromolecules according to their sedimentation properties (size, weight) in a density gradient.
